1580 The role of dendritic cell-regulatory T cell crosstalk in human head and neck cancer

We previously reported that Foxp3+CD4+ regulatory (Treg) cells expressing CTLA-4 on the cell surface are enriched in tumor microenvironment of human head and neck squamous carcinoma (HNSCC). Since is a key molecule to suppress dendritic (DCs), we investigated DCs HNSCC. Using scRNA-seq database, found HNSCC contained DC1, DC2 third DC subset expressed LAMP3 highly with migratory mature phenotype. The signature genes strongly correlated surface-CTLA4+Treg also observed LAMP3+DCs closely located Treg by immune fluorescence staining. both IL23A IL12B, which form IL-23 induce IL-17. Notably, higher expression IL17A, IL17F, or associated favorable prognosis These implicate novel role DC-Treg crosstalk contributes balancing IL-17 producing T cells.
